AminoSPARK nanoparticles and VivoTag fluorochromes enable development of novel  superbright fluorescence agents with properties that are ideal for extended use in in vivo or in vitro imaging.
Excellent for in vitro and in vivo imaging
IVM, FMT, FRI or fluorescence microscopy
Provide superbright, stable fluorescence for longer times than previously possible
Ideal for in vivo imaging
minimal cytotoxicity, narrow size distribution, highly functionalized
Customizable targeting
with amine-reactive molecules, such as NHS esters, anhydrides or EDC activated carboxylates
with chosen fluorochrome
Minimal cytotoxicity
Minimal cytotoxicity
Viability of RAW 264 cells incubated with nanoparticles for 24 hrs (assessed using CellTiter-Glo Luminescent Cell Viability Assay).
Dose-dependent uptake of nanoparticles in cytoplasm and nucleus
FACS analysis of labeled RAW 264 cells
FACS analysis of labeled RAW 264 cells after incubation with AminoSPARK 680 (1X and 10X concentration), derivatized with VivoTag 680, for 4 hrs and dual-channel fluorescence microscopy

Application examples

Arthritic mouse model: AminoSPARK 680 derivatized with VivoTag 680

Male BALB/c mice (5 weeks old)
A: Healthy, control
Male BALB/c mice (5 weeks old)
B: Arthritis -induced (intravenous injection with a collagen cocktail (4 mg/mouse, Chemicon), day 0, and intraperitoneal injection of lipopolysaccharide, (50 μg/ mouse), day 3
Male BALB/c mice (5 weeks old)
C: Dexamethasone-treated
Images: Male BALB/c mice (5 weeks old)
Investigation: Disease severity peaked at 8-9 days. Mice were injected intravenously with AminoSPARK 680 on day 7 and imaging took place after 24 hours.
