Quantification of therapeutic response in a model for human colorectal cancer

Investigation: Six to eight week-old female Nu/Nu mice implanted with A-673 human colorectal tumor cells. Treatment with Avastin started 7 days later. On Day 1 and Day 6 after treatment began, mice were injected with IntegriSense 680 and AngioSense 750 and imaged 20 hours later using FMT 2500 Imaging System.

Quantifying ectopic bone growth in vivo (OsteoSense)

Control and treated C3H/HeN mice.
Investigation: Control animals injected with LacZ cells, experimental animals injected with mesenchymal stem cells engineered to express the osteogenic recombinant human bone morphogenic protein 2 (rhBMP2).  OsteoSense 680 was injected on days 6 and 20. OsteoSense 750 was injected on day 13. In each case, imaging took place after 24 hours.

Imaging arthritic bone erosion (OsteoSense)

Quantitative fluorescence: Target-to-background ratio = specific fluorescence signal (relative fluorescence units) / adjacent tissue background signal. Statistical analysis: two-tailed unpaired Student t test.
Arthritic and healthy (non-induced) mice with regions of interest drawn within the paws. Arthritis was induced in DBA/1J mice (n=5 per group) by injection of bovine collagen, followed by a booster injection 21 days later. OsteoSense 680 was injected and imaging took place after 24 hours. Paw thickness was measured at the medial-lateral axis of the ankle joint.
